The NanoBRET® assay is a bioluminescence resonance energy transfer (BRET)-based assay that uses NanoLuc® Luciferase as the BRET energy donor and HaloTag® protein labeled with the HaloTag® NanoBRET® 618 fluorescent Ligand as the energy acceptor to measure the interaction of two binding partners in live cells.
The NanoBRET® Nano-Glo® Detection System provides the NanoBRET® Nano-Glo® Substrate used by NanoLuc® Luciferase to generate the donor signal and the HaloTag® NanoBRET® 618 Ligand for the fluorescent energy acceptor. The HaloTag® NanoBRET® 618 Ligand is added directly to the cells during plating, and the NanoBRET® Nano-Glo® Substrate is added to the sample just prior to measuring donor and acceptor emission. The NanoBRET® Nano-Glo® Kinetic Detection System has the Vivazine® substrate for use by the NanoLuc® luciferase donor, giving an extended signal for live-cell kinetic assays.
