Measure Compound Affinity and Occupancy at RAF Dimers in Live Cells​

RAF kinases (ARAF, BRAF and CRAF) play an important role in the MAPK signaling pathway, one of the most heavily mutated pathways in cancers. This signaling is typically mediated via the formation of RAF dimers, making these dimers important targets for drug discovery. The NanoBRET® TE Intracellular RAF Dimer Assays can be used to quantify compound affinity and fractional occupancy against RAF kinases in live cells. The assays rely on the NanoBRET® TE technology, a bioluminescence resonance energy transfer (BRET) method that uses a luciferase as the energy donor and a cell-permeable NanoBRET® TE Tracer as the energy acceptor. Instead of using NanoLuc® luciferase-fused target protein as the BRET donor, the NanoBRET® TE Intracellular RAF Dimer Assays use the NanoLuc® Binary Technology (NanoBiT) to enable the measurement of cellular target engagement at RAF dimers.

How the NanoBRET® TE RAF Dimer Assays Work

The NanoBRET® TE Intracellular RAF Dimer Assays use NanoLuc® Binary Technology (NanoBiT), a structural complementation system comprised of a large luciferase subunit (LgBiT) and a small complementary peptide (SmBiT(q)), to evaluate the target engagement of RAF kinases in dimeric complexes. Specifically, a fusion protein of LgBiT and full-length RAF is co-expressed with a fusion protein of SmBiT(q) and full-length RAF in cells. These are co-expressed with the constitutively active mutant KRAS(G12C). When the RAF proteins interact and form dimers in the presence of KRAS(G12C), the LgBiT and SmBiT(q) subunits come together to form an active luciferase enzyme that serves as the BRET donor. The energy acceptor is a cell-permeable fluorescent NanoBRET® tracer that binds specifically to one or both RAF protomers within the complex. Addition of compounds that bind to RAF protomer(s) displaces the tracer and results in a dose-dependent loss of BRET signal.

The NanoBiT® system used in the NanoBRET® TE Intracellular RAF Dimer Assays is unique in the use of the SmBiT(q) peptide, which makes the NanoBiT® luciferase enzyme sensitive to the Extracellular NanoLuc® Inhibitor. This inhibitor quenches signal that can arise from compromised cells and cellular debris to ensure the NanoBRET® signal originates from live cells.

Evaluate Target Engagement at RAF Homodimers and RAF Protomers in RAF Heterodimers​

Pairing the appropriate RAF fusion vectors with the recommended assay enables measurement of compound binding to BRAF and CRAF homodimers or to ARAF, BRAF and CRAF protomer within a RAF heterodimeric complex. To evaluate RAF homodimers, transfect cells with LgBiT and SmBiT(q) fusion vectors that express the same RAF protein of interest (Panels A–B and Table). To evaluate a specific RAF protomer within a RAF dimer, the RAF protomer of interest is paired with a second mutant RAF protein (Panels C–E and Table). The mutant RAF proteins contain an alanine-to-phenylalanine mutation that prevent tracer binding to the second RAF protomer but maintain the ability to dimerize with the RAF protomer of interest.

KRAS(G12C) Expression Vector is used with all target assays to drive RAF dimer formation.

Compound activity across multiple RAF configurations. The NanoBRET® TE Intracellular RAF Dimer Assays can be used to evaluate test compound activity across multiple RAF configurations using a similar workflow. Shown here are examples of activity profiles for RAF dimer inhibitor TAK-632 (Panel A) and LXH254 (Panel B). Both inhibitors can engage all RAF dimers, but both compounds show weaker affinity for CRAF within homodimers and for ARAF protomer.

Protomer selectivity of type II RAF inhibitors within the RAS/RAF complex

Next-generation RAF inhibitors that inhibit both RAF protomers within a RAF dimer can serve as therapeutics for RAF- and RAS-driven cancers. In this study published in Cell Chemical Biology, NanoBRET® Target Engagement assays were used to quantify target occupancy of RAF dimer inhibitors at selected RAF protomers within an active RAS-RAF complex in live cells. The results show that all inhibitors display reduced affinity for ARAF protomer in live cells. Moreover, ARAF protomer engagement, but not CRAF or BRAF engagement, corresponds with inhibition of RAS-RAF-mediated signaling pathways.

(2b) use NanoBRET®-branded energy acceptors (e.g., NanoBRET® tracers, NanoBRET® dyes, BRET-optimized HaloTag® ligands) for all determinations of energy transfer activity; or
(2c) contact Promega to obtain a license for use of the product for energy transfer assays using energy acceptors not manufactured by Promega. No license is needed if the energy transfer acceptor is a genetically encoded autofluorescent protein.

With respect to any uses outside this label license, including any diagnostic, therapeutic, prophylactic or commercial uses, please contact Promega for supply and licensing information.
