Measure Kinase-Compound Affinity in Live Cells
The NanoBRET® Target Engagement (TE) Intracellular Kinase Assays are based on the NanoBRET® System, an energy transfer technique designed to measure molecular proximity in living cells. The NanoBRET® TE Assays measure the apparent affinity of test compounds by competitive displacement of the NanoBRET® tracer, reversibly bound to a NanoLuc® luciferase-kinase fusion expressed in cells.

Extracellular techniques to measuring binding of kinase inhibitors can fail to accurately predict intracellular target engagement. In this article, published in Cell Chemical Biology, we report the application of an energy transfer technique that enables the first equilibrium-based approach to quantitatively profile target occupancy and compound affinity in live cells.
Discovery of a Selective Inhibitor of Doublecortin-Like Kinase 1
Doublecortin-like kinase 1 (DCLK1) is an understudied kinase that is upregulated in a wide range of cancers, including pancreatic ductal adenocarcinoma (PDAC). This study uses NanoBRET® technology to understand the function of DCLK1 in PDAC cells.
Quantifying CDK Inhibitor Selectivity in Live Cells
Teams at the Structural Genomics Consortium and Promega used the NanoBRET® Target Engagement technology to uncover surprising patterns of selectivity for touted CDKIs and abandoned clinical leads.
